Le mercredi 21 février 2024, Madame Noémie BRICMONT, titulaire d'un Master en Sciences biomédicales à finalité approfondie et d’un Certificat de formation à la recherche en sciences biomédicales et pharmaceutiques, présentera l'examen en vue de l'obtention du grade de Doctorat en sciences biomédicales et pharmaceutiques, sous la direction de Madame Céline KEMPENEERS et de Madame Marie-Christine SEGHAYE.

Cette épreuve consistera en la défense publique d'une thèse intitulée :

«Standardisation and validation of ciliary videomicroscopy using nasal respiratory epithelium samples»

Résumé de la thèse

Primary ciliary dyskinesia (PCD) is a genetic motile ciliopathy, in which respiratory cilia beat abnormally, leading to impaired mucociliary clearance and chronic otosinopulmonary disease.  

Currently, no single test will diagnose every patient with PCD, and the diagnosis involves a panel of 5 different diagnostic tests. According to the European Respiratory Society guidelines, hallmark ultrastructural defects by transmission electron microscopy and bi-allelic mutations in PCD causing genes can confirm a PCD diagnosis, but these tests will miss 15-30% of patients.  Ciliary beating can be evaluated using digital high-speed videomicroscopy (DHSV), a highly sensitive and specific test for PCD diagnosis, but international recommendations stated that currently, DHSV is not sufficiently standardized (lack of published formal operating procedures for processing and analyzing samples using DHSV) to confirm a PCD diagnosis.  
Indeed, the experimental conditions may influence ciliary function, and given the lack of evidence-based data and formal recommendations for the optimal respiratory sample collection and processing, various laboratories use differing protocols, and consequently have different reference values. This research project focuses on the influence of the respiratory sample storage conditions and the standard operation procedure to perform ciliary functional analysis, on ciliary beating evaluation
