Cette épreuve consistera en la défense publique d'un mémoire intitulé :

"Role of the SV2A protein in parvalbumin expressing GABAergic interneurons in the epileptic disease physiopathology"

SV2A is a glycoprotein present in the membranes of most synaptic vesicles whose physiological role remains unknown. Interestingly, it has been demonstrated that an effective anti-epileptic drug named Levetiracetam binds to SV2A. Knowing that Epilepsy is one of the most common neurological disease and that around twenty-five percent of epileptic patient cannot be treated, we wanted to decipher the link between SV2A and Epilepsy using a recombinant animal model. This work identifies parvalbumin expressing GABAergic interneurons as the main cellular actor in the relationship between SV2A and epilepsy. The presence of SV2A in those neurons is also necessary for the anti-epileptic activity of the Levetiracetam. In addition, our results demonstrate that the hippocampus is particularly affected by SV2A absence in inhibitory interneurons or parvalbumin expressing interneurons only. At least so far, our hypothesis concerning the role of SV2A in those interneurons would imply that the recycling role of Syt1 performed by SV2A weight more heavily on parvalbumin expressing neurons compare to other sub-types of neurons or interneurons.
